JAK-STAT Signaling
Many cytokines and growth factors communicate with cells through receptors associated with JAK enzymes.
Activation of JAK1 and JAK2 can lead to phosphorylation and activation of STAT proteins. Activated STAT proteins move into the cell nucleus and influence gene expression.
Ruxolitinib interferes with this signaling process by inhibiting JAK1 and JAK2.
Topical Ruxolitinib
With Rutinib Cream, ruxolitinib is delivered to the skin.
This formulation is designed for topical treatment of specified dermatologic diseases. The clinical effects relate to local skin inflammation and immune signaling, although topical treatment can still produce systemic exposure.
Oral Ruxolitinib
Rutinib 5 mg and Ruxotor 10 mg represent oral ruxolitinib products.
Oral administration produces systemic exposure and is used in systemic disease settings such as myelofibrosis, polycythemia vera and graft-versus-host disease under applicable labeling.
Why Formulation Matters
The same active ingredient can have different clinical effects depending on:
- route
- concentration
- dose
- absorption
- systemic exposure
- treatment area
- patient population
- disease being treated
- regulatory authorization
Therefore, topical and oral ruxolitinib should not be considered equivalent treatments.
